Introduction-                   

Deflazacort is a Glucocorticoid (glucose + cortex+ steroid). It regulates the conversion of fats, proteins, and carbohydrates to energy. It also helps to regulate blood pressure & cardiovascular function. It has the following actions

1.    Anti-inflammatory 

2.    Immunosuppressant

Class-  

It belongs to the class of Corticosteroids (Glucocorticoid).

Mechanism of action   

Corticosteroid inhibits mRNA responsible for interleukin- 1formation.Interleukins produces anti-inflammatory, immunosuppressant effects. The steroid hormone binds to a specific receptor in the cytoplasm. Steroids work by decreasing the release of chemical substance thereby reducing inflammation so that the immune system activity is reduced.

Pharmacokinetic-  Protein binding of deflazacort is about 40%. 

                                Half-life (t1/2) of deflazacort is 1.1-1.9 hours

                                Elimination of deflazacort -Urinary Excretion

Pharmacological Uses-       

Deflazacort can be prescribed in these indications- Autoimmune diseases (e.g. - systemic lupus erythematosus, autoimmune hepatitis), Joint & Muscle diseases (e.g.-Rheumatoid arthritis), Allergies, Asthma, Cancer, Inflammation of skin, Nephrotic syndrome, Ulcerative colitis, Autoimmune hemolytic anaemia, Hypersensitivity reaction, Crohn's disease, Anaphylaxis & Ulcerative colitis.

Side Effects of Deflazacort

Skin lesions (like- acne, bruises, stretch market etc.)

Recurrent infection (like- the flu, a cold, pneumonia) 

Muscle or bone weakness (osteoporosis)                                                              

Cushing’s syndrome 

Infrequent or absent menstrual periods

Hirsutism

Increased appetite

Tablet- 6mg, 18mg, 30mg, 36mg.                                                                                          

Oral suspension- 22.75mg/mL
