Introduction

Amlodipine Besylate is a part of the family calcium channel blocker for lowering the blood pressure in hypertension. They work to lessen the workload (pump) of Heart by relaxing the blood vessels & are a long-acting product. It increases the input of blood to the heart to suppress the chest pain.

Mechanism of Action

Amlodipine works on the principle of blocking the calcium ion influx across the cell membrane on vascular smooth muscle cells with a greater effect as compared to cardiac muscle cells. As a result, vasodilatation occurs & peripheral vascular resistance decreases to lower the blood pressure.

Pharmacological Uses 

Amlodipine is used for the management of high blood pressure, coronary artery disease, Heart-related chest pain, chronic stable angina, Vasospastic angina, Prevent Strokes.

There are a number of products which can interact with the Amlodipine 

Alprenolol, Acebutolol (its hypotensive action increases)

Alfuzosin, Amphetamine (increases hypotensive action)

Amphotericin B, Amorolfine (adverse effects)

Barbexaclone, Barbital (Increases metabolism)

Dosage

Starting dose-5mg OD (once a day)

Maintaining does-5-10mg OD (once a day)

The dose should not exceed 10mg per day. For the fragile patient, 2.5mg is enough.
